Custom DNA Synthesis
PrimeSyn Lab offers over 15 years of expertise in the area of custom DNA synthesis, primers, and modified oligonucleotide synthesis. Custom oligonucleotides are synthesized on ABI 3900, and expedite 8909. Custom DNA primers can be ordered from PSL in a different scales and purity level. PSL uses its proprietary developed methodology to synthesize and purify custom DNA primers. These custom DNA primers can be used in a variety of applications from PCR, diagnostic, and fluorescent labeled probes. Custom DNA synthesis includes:
- Unmodified primers
- Proprietary methodology in making poly “G” sequences and their purifications

Quality Control of custom DNA and Primers (HPLC, Mass Spectral analysis and Capillary Electrophoresis):
PrimeSyn Lab Inc. has proprietary HPLC purification methodology whereby it is possible to achieve very difficult separation in case of modified oligonucleotides and sequences containing multiple "G" and "C" bases. Our final product undergoes stringent testing before shipping. We have ion-exchange and reverse-phase methodologies which enables us to even separate sequences containing wobble bases. PrimeSyn Lab also uses Capillary Electrophoresis (P/ACE™ MDQ ) , which is an excellent analytical technique with high sensitivity, high resolution, and has a robust ability for determining the purity of the DNA.
Each custom DNA synthesized by PSL undergoes stringent quality control such as HPLC, and mass spectral analysis. PSL offers complete guarantee for synthesized unmodified primers or other custom DNA to exceed the customer’s satisfaction. We stand behind our product and customers can ask for product replacement within 30 days if they are not completely satisfied with the custom primers or any other products. A comprehensive certificate of analysis provided with each custom DNA or primers includes information such as: sequence, purchase order number, lot number, concentration, and the total number of OD. PSL can make custom label based on their clients needs. PSL Confirms molecular mass ( +/- 5 amu) for synthetic oligomers up to 100 mer which helps us to monitor effieciency of synthesis and capping of each custom DNA primers. The importance of mass spectroscopy in the quality control of custom DNA and primers are proven by the following example. In one such synthesis, we observed multiple +28 peak due to the N-formyl group, a result of incomplete cleavage of dimethylformamidine group on "G" bases in the presence of ammonium hydroxide at room temperature. Interestingly, both HPLC and capillary electrophoresis indicated one peak. Re-suspension in 30% ammonium hydroxide at 55 oC for two hours resulted in complete disappearance of the +28 amu peak.
